🧬 Genetic Drug Sensitivities

🔴 CYP2B11 (Reduced hepatic metabolism)

Prevalence: Unknown

Sighthound anesthetic sensitivity. Low body fat and reduced CYP2B11 activity. Reduce induction dose by 25-50%.

Simparica Trio contains moxidectin (0.024-0.048 mg/kg), a macrocyclic lactone and P-glycoprotein substrate. In MDR1-mutant dogs, moxidectin accumulates in CNS causing dose-dependent neurotoxicity. FDA label warns against use in MDR1-affected breeds.

AVOIDSimparica Trio→ Alternatives: sarolaner alone (Simparica — no moxidectin), afoxolaner (NexGard) for fleas/ticks + selamectin for HW prevention

Also avoid: Simparica Trio, moxidectin, ivermectin

Sarolaner alone (Simparica, not Trio) is safe in MDR1 breeds

AVOIDIvermectin→ Alternatives: selamectin, spinosad, afoxolaner, lotilaner

Also avoid: ivermectin, milbemycin oxime, moxidectin, loperamide

Test for MDR1 mutation before prescribing macrocyclic lactones in herding breeds (WSU MDR1 test available)

Use Interceptor (milbemycin) with caution — also MDR1-substrate at higher doses; preventive doses generally safe but verify

🏥 Anesthesia Considerations

⚠ Sighthound Anesthetic Sensitivity

Low body fat → prolonged recovery from lipid-soluble agents (thiopental, propofol). Reduce induction doses 25-50%. Use lean-body-mass dosing. CYP2B11 deficiency in Greyhounds = slow barbiturate/propofol metabolism. Prefer alfaxalone for induction. Extended post-anesthetic monitoring required.
